Drug Combo Offers Long-Term Benefits in SLE

WASHINGTON -- An 18-month treatment with the combination of rituximab and cyclophosphamide (Cytoxan) stopped progression of systemic lupus erythematosus (SLE) for at least 3 years, researchers here.

The mean SLE Disease Activity Index (SLEDAI) score decreased from 8.929 to 1.917 after the first 6 months of therapy and remained low thereafter, reported Thomas Lehman, MD, from the Hospital for Special Surgery in New York City, and colleagues at the at American College of Rheumatology meeting.

He added that there also were significant drops in prednisone use at various time points, from 29 mg/day at baseline to (all P<0.001):

1 year: 14.74 mg/day

2 years: 10.43 mg/day

3 years: 8.4 mg/day

At 5 years, corticosteroid use was at 8.5 mg/day (P<0.01).

All of 15 patients in the study had active SLE despite prior therapy. They received rituximab (Rituxan) at a dose of 750 mg/m2 on day 1 and cyclophosphamide (Cytoxan) at a dose of 750 mg/m2 on day 2.
